At the moment RDM does not support ED25519 on Yubikey. We support RSA and ECDSA. That being said, it seems that your question is more "Can I use the key stocked inside the Yubikey in the key agent". If this is what you've meant, yes, if you are able to extract the private key from the Yubikey, it can then be used in the key agent.
