Yesterday I tried to reconfigure the SQL Data Source for a new user but I was not able to connect to the database or the Host ! I’ve got the popup error:" Unable to connect to the server!" So I tried to connect with a different user this morning and it did not work.

Which version of RDM are you currently using?
Also, have you created these users under Administration -> Users in RDM first?
