we have 2 user out of 15 total that cannot configure the central mssql datastore in rdm 10.1.7.0 anymore without local administration rights.
There are no entries visible in the rdm help log or in the local eventvwr. As soon as the user gets local admin rights, this is working again.

Could you verify the error logs in Help->View Application Log
Do they get an error? You could also try Ctrl+Refresh button to clear the cache

There is no entry in the Application Log as rdm crashes as soon as it trying to access the sql datastore. When I logon
on his local computer with my account and connect to the datastore, this is working without any issues. So i believe
it is a local client issue. This was running perfectly until yesterday.
We then had to install the latest version of vmware horizon 3.2.0 client and the vmware client integration plug-in 5.5.0. This was the last change on that machine. Could this be a possible conflict??

We just tried this, same effect. RDM crashes upon loading sessions. It looks like as the client doesn't even try to open a connection to the sql server. (at least according to netstat -ano)

We finally had to completely restage the affected machine, now it working again. I believe there is / was an incompatibility between rdm and a piece of software :(
