I'd like to use the Remote Desktop Manager as portable app. Is there a way to use the Connections.mdb from an other path as this one in the user profile ?

Yes it's possible, install RDM on an usb drive, and copy the mdb on it. After that start the application from the mdb and change the data source path to only the mdb name (no path).

This worked pretty fine, but now I activated the Autostartfunction and then after a system reboot I get an error message that the currect database is unavailable. RDM searches the database in "c:\Windows\SysWOW64\Connections.mdb".
When I close the automatically started RDM and start it manually it work's pretty fine.

It seems that the working directory is by default c:\Windows\SysWOW64\ when it executed by Windows, I will try to force it to the installation path. Can you try this version:
